The Nitration of Proteins, Lipids and DNA by Peroxynitrite Derivatives-Chemistry Involved and Biological Relevance

In recent years, much interest has been generated by the idea that nitrosative stress plays a role in aetiology of human diseases, such as atherosclerosis, inflammation, cancer, and neurological diseases. The chemical changes mediated reactive nitrogen species (RNS) are detrimental to cell function, because they can cause nitration, which alter structures cellular proteins, DNA, lipids, hence, impair their normal function. One most potent biological agents is peroxynitrite (ONOO?), produced when nitric oxide (•NO) superoxide (•O2?) combined at extremely rapid rates. Considering plethora oxidations peroxynitrite, this makes prevalent nitrating responsible for protein, lipids nitration vivo. There biochemical evidence suggest interactions radicals NO result formation redox system, includes reactions nitrosation component complex signalling network. However, chemistry involved process with derivatives poorly understood, particularly molecules, lipids. Here, we review processes biomolecules, provide mechanistic explanation NOS stress. This study reveals these based on surprisingly simple straightforward chemistry, fascinating influence physiology pathology.
